Customizing Your Origin/Optional Class Features: All TCoE rules for customizing your origin and optional class features are available.

Diagonals: When moving diagonally on a grid, the first diagonal move costs 5 feet, and the second costs 10 feet; the pattern continues to alternate between 5 and 10 feet throughout any diagonal movement on a turn, such that moving two squares diagonally, whether consecutively or not, always costs 15 feet.

Inspiration: You can spend inspiration to gain advantage on an ability check, attack roll, or saving throw. You either have inspiration or you don't; you can't stockpile it.

Mixing Potions: Consuming multiple potions might have some unpredictable effects.

Potion Consumption: It takes a bonus action to consume a potion, but administering a potion to an unconscious creature takes an action.
Setting-Specific Content: Subclasses, like the oath of glory paladin from the Theros setting, are generally permitted. Backgrounds, like those from the Strixhaven setting, are generally not permitted. Exceptions may be considered on an individual basis.
